L606M : Biochemical assays
The L606M substitution in human POLD1 and the analogous L612M substitution in Saccharomyces cerevisiae Pol3 increase the polymerase activity of DNA polymerase δ and decrease the accuracy of DNA synthesis in vitro. Data with yeast L612M variant indicates a defect in polymerase-to-exonuclease active site switching.
